The Correspondence Study of BIAcore Binding and Anti-HBV Effect in Vitro by KA-01 and Its Main Ingredient

    • In order to discover activated ingredient form Chinese herbal medicine and to bind KA-01and its main ingredients to HBsAg and HBeAg protein in CM5 by BIAcore 3000,the HepG2.2.15 cell was cultured in vitro to observe the inhibitory effect of KA-01 and its main ingredient. The inhibitory effect of KA-01 and its main ingredients on protein secretion of HBV were tested by Laser Confocal Microscope. The results indicated that the value of KA-01and its main ingredients of A、B、C、D binding with HBsAg protein were 200、183.7、162.1、39.1、66.6 RU respectively, but almost 0 with HBeAg protein. The inhibitory effect of KA-01 on secretion of HBsAg and HBeAg were 62.01% and 30.59% respectively. The inhibitory effect of A on secretion of HBsAg and HBeAg were 54.26% and 14.15% respectively. The inhibitory effect of B on secretion of HBsAg and HBeAg were 50.97% and 27.92% respectively. The inhibitory effect of C on secretion of HBsAg and HBeAg were 17.54% and 8.06% respectively. The conclusion is that KA-01and its main ingredients of A、B have the inhibitory effect on HBV.
