Analysis and Succession of Microbial Community Structure in Hybrid MBR for Landfill Leachate Treatment

摘要: 为了揭示复合式膜生物反应器中微生物群落结构多样性的演变过程,为改进工艺提供依据,分别3次从反应器混合液中和生物膜上采集样品并提取样品的微生物总DNA, 然后使用V6-V8区引物对(GC968F/1492R)进行PCR扩增,最后用PCR扩增产物进行变性梯度凝胶电泳(DGGE).DGGE分析表明,在反应器运行初期微生物群落结构变化较大;在整个反应器运行过程中,微生物群落多样性较高;在该系统中,由于有生物填料的存在,可能存在厌氧微环境,所以在该反应器中可能存在短程脱氮和同步脱氮.在反应器运行约2个月时,出现的一些菌群是膜污染物产生和累积的主要因素.Abstract: In order to reveal the succession of microbial community diversity in a hybrid membrane bioreactor (MBR) and provide microbial evidence for technique improvements,total microbial DNA is extracted from six samples which are collected three times respectively from mixed liquor and biofilm of a MBR,16SrDNA fragments were amplified from the total DNA successfully using a pair of universal bacterial 6SrDNA primer,GC968F and 1492R,and then the amplified PCR are used for denaturing gradient gel electrophoresis (DGGE) analysis.DGGE analysis indicated that a noticeable change takes place in microbial structure at initial stages of MBR operation.In the whole operation,bacterial community structures are higher.In this bioreactor,because of the existence of filling,there might be two different modes of enitrification in the reactor because there might be anaerobic environment in it.Two months later,some increasing bacteria has greater impact on the production and accumulation of membrane pollutants.
